@article{fdi:010067733, title = {{B}acteria of the genus {R}hodopseudomonas ({B}radyrhizobiaceae) : obligate symbionts in mycelial cultures of the black truffles {T}uber melanosporum and {T}uber brumale}, author = {{L}e {R}oux, {C}. and {T}ournier, {E}. and {L}ies, {A}. and {S}anguin, {H}. and {C}hevalier, {G}. and {D}uponnois, {R}obin and {M}ousain, {D}. and {P}rin, {Y}.}, editor = {}, language = {{ENG}}, abstract = {{B}ackground: {T}his work aimed at characterizing 12 isolates of the genus {T}uber including {T}uber melanosporum (11 isolates) and {T}uber brumale (one isolate). {T}his was done using internal transcribed spacer ({ITS}) sequences, confirming their origin. {R}esults: {A}nalysis of their mating type revealed that both {MAT}1-1 and {MAT}1-2 exist within these isolates (with 3 and 8 of each, respectively). {W}e observed that each of these cultures was consistently associated with one bacterium that was intimately linked to fungal growth. {T}hese bacterial associates failed to grow in the absence of fungus. {W}e extracted {DNA} from bacterial colonies in the margin of mycelium and sequenced a nearly complete 16{S} r{DNA} gene and a partial {ITS} fragment. {W}e found they all belonged to the genus {R}hodopseudomonas, fitting within different phylogenetic clusters. {N}o relationships were evidenced between bacterial and fungal strains or mating types. {R}hodopseudomonas being a sister genus to {B}radyrhizobium, we tested the nodulation ability of these bacteria on a promiscuously nodulating legume ({A}cacia mangium), without success. {W}e failed to identify any nif{H} genes among these isolates, using two different sets of primers. {C}onclusions: {W}hile the mechanisms of interaction between {T}uber and {R}hodopseudomonas remain to be elucidated, their interdependency for in vitro growth seems a novel feature of this fungus.}, keywords = {{T}ruffle ; {A}scomycete ; {P}rocaryote ; {I}n vitro production ; {C}ultivability}, booktitle = {}, journal = {{S}pringerplus}, volume = {5}, numero = {}, pages = {art. 1085 [10 p.]}, ISSN = {2193-1801}, year = {2016}, DOI = {10.1186/s40064-016-2756-6}, URL = {https://www.documentation.ird.fr/hor/fdi:010067733}, }
