@article{fdi:010066843, title = {{P}rokaryotic diversity in a {T}unisian hypersaline lake, {C}hott {E}l {J}erid}, author = {{B}en {A}bdallah, {M}. and {K}array, {F}. and {M}hiri, {N}. and {M}ei, {N}. and {Q}u{\'e}m{\'e}neur, {M}arianne and {C}ayol, {J}. {L}. and {E}rauso, {G}. and {T}holozan, {J}ean-{L}uc and {A}lazard, {D}idier and {S}ayadi, {S}.}, editor = {}, language = {{ENG}}, abstract = {{P}rokaryotic diversity was investigated in a {T}unisian salt lake, {C}hott {E}l {J}erid, by quantitative real-time {PCR}, denaturing gradient gel electrophoresis ({DGGE}) fingerprinting methods targeting the 16{S} r{RNA} gene and culture-dependent methods. {T}wo different samples {S}1-10 and {S}2-10 were taken from under the salt crust of {C}hott {E}l {J}erid in the dry season. {DGGE} analysis revealed that bacterial sequences were related to {F}irmicutes, {P}roteobacteria, unclassified bacteria, and {D}einococcus-{T}hermus phyla. {A}naerobic fermentative and sulfate-reducing bacteria were also detected in this ecosystem. {W}ithin the domain archaea, all sequences were affiliated to {E}uryarchaeota phylum. {Q}uantitative real-time {PCR} showed that 16{S} r{RNA} gene copy numbers of bacteria was 5 x 10(6) {DNA} copies g(-1) whereas archaea varied between 5 x 10(5) and 10(6) {DNA} copies g(-1) in these samples. {E}ight anaerobic halophilic fermentative bacterial strains were isolated and affiliated with the species {H}alanaerobium alcaliphilum, {H}alanaerobium saccharolyticum, and {S}porohalobacter salinus. {T}hese data showed an abundant and diverse microbial community detected in the hypersaline thalassohaline environment of {C}hott {E}l {J}erid.}, keywords = {{A}rchaea ; {B}acteria ; {M}icrobial diversity ; {H}ypersaline lake ; 16{S} r{RNA} ; {PCR}-{DGGE} ; {Q}uantitative {PCR} ; {A}naerobic fermentative bacteria ; {TUNISIE}}, booktitle = {}, journal = {{E}xtremophiles}, volume = {20}, numero = {2}, pages = {125--138}, ISSN = {1431-0651}, year = {2016}, DOI = {10.1007/s00792-015-0805-7}, URL = {https://www.documentation.ird.fr/hor/fdi:010066843}, }
