@article{fdi:010066119, title = {{C}omplete genome sequence and description of {S}alinispira pacifica gen. nov., sp. nov., a novel spirochaete isolated form a hypersaline microbial mat}, author = {{B}en {H}ania, {W}ajdi and {J}oseph, {M}anon and {S}chumann, {P}. and {B}unk, {B}. and {F}iebig, {A}. and {S}proer, {C}. and {K}lenk, {H}. {P}. and {F}ardeau, {M}arie-{L}aure and {S}pring, {S}.}, editor = {}, language = {{ENG}}, abstract = {{D}uring a study of the anaerobic microbial community of a lithifying hypersaline microbial mat of {L}ake 21 on the {K}iritimati atoll ({K}iribati {R}epublic, {C}entral {P}acific) strain {L}21-{RP}ul-{D}2({T}) was isolated. {T}he closest phylogenetic neighbor was {S}pirochaeta africana {Z}-7692({T}) that shared a 16{S} r{RNA} gene sequence identity value of 90% with the novel strain and thus was only distantly related. {A} comprehensive polyphasic study including determination of the complete genome sequence was initiated to characterize the novel isolate. {C}ells of strain {L}21-{RP}ul-{D}2({T}) had a size of 0.2 - 0.25 x 8-9 mu m, were helical, motile, stained {G}ram-negative and produced an orange carotenoid-like pigment. {O}ptimal conditions for growth were 35 degrees {C}, a salinity of 50 g/l {N}a{C}l and a p{H} around 7.0. {P}referred substrates for growth were carbohydrates and a few carboxylic acids. {T}he novel strain had an obligate fermentative metabolism and produced ethanol, acetate, lactate, hydrogen and carbon dioxide during growth on glucose. {S}train {L}21-{RP}ul-{D}2({T}) was aerotolerant, but oxygen did not stimulate growth. {M}ajor cellular fatty acids were {C}-14:0, iso-{C}-15:0, {C}-16:0 and {C}-18:0. {T}he major polar lipids were an unidentified aminolipid, phosphatidylglycerol, an unidentified phospholipid and two unidentified glycolipids. {W}hole cell hydrolysates contained {L}-ornithine as diagnostic diamino acid of the cell wall peptidoglycan. {T}he complete genome sequence was determined and annotated. {T}he genome comprised one circular chromosome with a size of 3.78 {M}bp that contained 3450 protein-coding genes and 50 {RNA} genes, including 2 operons of ribosomal {RNA} genes. {T}he {DNA} {G} + {C} content was determined from the genome sequence as 51.9 mol%. {T}here were no predicted genes encoding cytochromes or enzymes responsible for the biosynthesis of respiratory lipoquinones. {B}ased on significant differences to the uncultured type species of the genus {S}pirochaeta, {S}. plicatilis, as well as to any other phylogenetically related cultured species it is suggested to place strain {L}21-{RP}ul-{D}2({T}) (={DSM} 27196({T}) = {JCM} 18663({T})) in a novel species and genus, for which the name {S}alinispira pacifica gen. nov., sp. nov. is proposed.}, keywords = {{S}pirochaetes ; {F}ermentative metabolism ; {O}xygen tolerance ; {H}ypersaline microbial mat ; {K}iritimati atoll ; {PACIFIQUE} ; {KIRIBATI} ; {KIRITIMATI} {ATOLL}}, booktitle = {}, journal = {{S}tandards in {G}enomic {S}ciences}, volume = {10}, numero = {}, pages = {art. 7 [14 p.]}, ISSN = {1944-3277}, year = {2015}, DOI = {10.1186/1944-3277-10-7}, URL = {https://www.documentation.ird.fr/hor/fdi:010066119}, }
