@article{fdi:010066040, title = {{D}etermination of backscattering cross section of individual particles from cytometric measurements : a new methodology}, author = {{D}uforet-{G}aurier, {L}. and {M}outier, {W}. and {G}uiselin, {N}. and {T}hyssen, {M}. and {D}ubelaar, {G}. and {M}eriaux, {X}. and {C}ourcot, {L}. and {D}essailly, {D}. and {L}oisel, {H}ubert}, editor = {}, language = {{ENG}}, abstract = {{A} methodology is developed to derive the backscattering cross section of individual particles as measured with the {C}yto{S}ense ({C}yto{B}uoy b.v., {NL}). {T}his in situ flow cytometer detects light scatter in forward and sideward directions and fluorescence in various spectral bands for a wide range of particles. {F}irst, the weighting functions are determined for the forward and sideward detectors to take into account their instrumental response as a function of the scattering angle. {T}he {C}yto{S}ense values are converted into forward and sideward scattering cross sections. {T}he {C}yto{S}ense estimates of uniform polystyrene microspheres from 1 to 90 m m are compared with {M}ie computations. {T}he mean absolute relative differences {D}elta {E} are around 33.7% and 23.9% for forward and sideward scattering, respectively. {T}hen, a theoretical relationship is developed to convert sideward scattering into backscattering cross section, from a synthetic database of 495,900 simulations including homogeneous and multi-layered spheres. {T}he relationship follows a power law with a coefficient of determination of 0.95. {T}o test the methodology, a laboratory experiment is carried out on a suspension of silica beads to compare backscattering cross section as measured by the {WET} {L}abs {ECO}-{BB}9 and derived from {C}yto{S}ense. {R}elative differences are between 35% and 60%. {T}hey are of the same order of magnitude as the instrumental variability. {D}ifferences can be partly explained by the fact that the two instruments do not measure exactly the same parameter: the cross section of individual particles for the {C}yto{S}ense and the bulk cross section for the {ECO}-{BB}9.}, keywords = {}, booktitle = {}, journal = {{O}ptics {E}xpress}, volume = {23}, numero = {24}, pages = {31510--31533}, ISSN = {1094-4087}, year = {2015}, DOI = {10.1364/oe.23.031510}, URL = {https://www.documentation.ird.fr/hor/fdi:010066040}, }
