@article{fdi:010064063, title = {{O}ptimization of the conditions for {C}asuarina cunninghamiana {M}iq. genetic transformation mediated by {A}grobacterium tumefaciens}, author = {{J}iang, {Q}. {B}. and {M}a, {Y}. {Z}. and {Z}hong, {C}. {L}. and {Z}eng, {B}. {S}. and {Z}hang, {Y}. and {P}inyopusarerk, {K}. and {B}ogusz, {D}idier and {F}ranche, {C}laudine}, editor = {}, language = {{ENG}}, abstract = {{U}sing epicotyl fragments of the actinorhizal tree {C}asuarina cunninghamiana and the disarmed strain of {A}grobacterium tumefaciens {C}58{C}1 (p{GV}2260) containing the p{BIN}19-35{S}-{GUSINT} binary vector, a method for the genetic transformation of {C}. cunninghamiana was established. {T}ransformed cells were initially selected for 2 weeks on nutrient medium supplemented with kanamycin 20 mg {L}-1 during callus induction, and then subcultured with 50 mg {L}-1 until adventitious bud and shoot differentiation. {D}ifferent factors involved in the early stages of the {T}-{DNA} transfer process were studied. {A}grobacterium-mediated {DNA} delivery was most successful when epicotyl fragments excised from 45-day-old seedlings were co-cultivated with an exponentially growing culture of {A}. tumefaciens at an {OD}600nm of 0.3, for 5 days in the presence of 50 mu {M} of acetosyringone. {K}anamycin resistant calli were observed on 88.89 % of the explants and transgenic rooted {C}. cunninghamiana plants were obtained in 6 months. {E}vidence of genetic transformation was demonstrated by -glucuronidase histochemical assays and polymerase chain reaction analyses. {T}he possibility to obtain transgenic nitrogen-fixing nodules after inoculation by the soil actinomycete {F}rankia was established.}, keywords = {{A}ctinorhizal plant ; {C}asuarina cunninghamiana ; {A}grobacterium tumefaciens ; {G}enetic transformation ; {F}rankia}, booktitle = {}, journal = {{P}lant {C}ell {T}issue and {O}rgan {C}ulture}, volume = {121}, numero = {1}, pages = {195--204}, ISSN = {0167-6857}, year = {2015}, DOI = {10.1007/s11240-014-0694-y}, URL = {https://www.documentation.ird.fr/hor/fdi:010064063}, }
