@article{fdi:010063980, title = {{D}evelopment and validation of liquid chromatography combined with tandem mass spectrometry methods for the quantitation of simalikalactone {E} in extracts of {Q}uassia amara {L}. and in mouse blood}, author = {{L}e, {H}. {L}. and {J}ullian, {V}al{\'e}rie and {C}laparols, {C}. and {V}ansteelandt, {M}. and {H}addad, {M}ohamed and {C}abou, {C}. and {D}eharo, {E}ric and {F}abre, {N}icolas}, editor = {}, language = {{ENG}}, abstract = {{I}ntroduction{S}imalikalactone {E} ({S}k{E}) from {Q}uassia amara, has been proved to be a valuable anti-malarial and anti-cancer compound. {A}s {S}k{E} is very scarce, methods of quantitation are needed in order to optimise its isolation process and to determine pharmacokinetic data. {O}bjective{T}o validate methods using liquid chromatography coupled to mass spectrometry for the quantitation of {S}k{E} in plant extracts and in biological fluids. {M}ethods{H}igh- and ultrahigh-performance liquid chromatography ({UHPLC}) coupled to ion trap mass spectrometry ({MS}) with single ion monitoring detection and to triple quadrupole-linear ion trap tandem mass spectrometry with multiple reaction monitoring detection methods were developed. {V}alidation procedure was realised according to the {I}nternational {C}onference on {H}armonisation guideline. {M}ethanol extracts of dried {Q}uassia amara leaves, and mouse-blood samples obtained after various routes of administration, were analysed for {S}k{E}. {R}esults{M}ethods were validated and gave similar results regarding the content of {S}k{E} expressed per kilogram of dry leaves in the traditional decoction (16012mg/kg) and in the methanol extract (932mg/kg). {T}he recovery of the analyte from mouse blood ranged from 80.7 to 119.8%. {S}imalikalactone {E} was only detected using {UHPLC}-{MS}/{MS} (0.2 +/- 0.03mg/{L}) in mouse blood after intravenous injection: none was detected following intraperitoneal or oral gavage administration of {S}k{E}. {C}onclusion{T}he {LC}-{MS} methods were used for the quantitation of {S}k{E} in plant extracts and in mouse blood. {T}hese methods open the way for further protocol optimisation of {S}k{E} extraction and the determination of its pharmacokinetic data.}, keywords = {{LC}-{MS} ; quantitation ; biological fluids ; plant extract ; quassinoids ; simalikalactone {E} ; {Q}uassia amara ; {GUYANE} {FRANCAISE}}, booktitle = {}, journal = {{P}hytochemical {A}nalysis}, volume = {26}, numero = {2}, pages = {111--118}, ISSN = {0958-0344}, year = {2015}, DOI = {10.1002/pca.2542}, URL = {https://www.documentation.ird.fr/hor/fdi:010063980}, }
