@article{fdi:010063913, title = {{C}onstruction of a genetic map for pearl millet, {P}ennisetum glaucum ({L}.) {R}. {B}r., using a genotyping-by-sequencing ({GBS}) approach}, author = {{M}oumouni, {K}. {H}. and {K}ountche, {B}. {A}. and {J}ean, {M}. and {H}ash, {C}. {T}. and {V}igouroux, {Y}ves and {H}aussmann, {B}. {I}. {G}. and {B}elzile, {F}.}, editor = {}, language = {{ENG}}, abstract = {{P}earl millet is the main component of traditional farming systems and a staple grain in the diet of sub-{S}aharan {A}frica and {I}ndia. {T}o facilitate breeding work in this crop, a genetic map consisting of single nucleotide polymorphism ({SNP}) markers was constructed using an {F}-2 population of 93 progenies, from a wild x cultivated pearl millet cross. {W}e used a modified genotyping-by-sequencing ({GBS}) protocol involving two restriction enzymes ({P}st{I}-{M}sp{I}) and {PCR} amplification with primers including three selective bases to generate 3,321 {SNP}s. {O}f these, 2,809 high-quality {SNP}s exhibited a minor allele frequency >= 0.3. {I}n total, 314 non-redundant haplotypes and 85 {F}-2 individuals were used to construct a genetic map spanning a total distance of 640 c{M}. {T}hese {SNP}s were evenly distributed over seven linkage groups ranging considerably in size (62-123 c{M}). {T}he average density for this map was 0.51 {SNP}/c{M}, and the average interval between {SNP} markers was 2.1 (+/- 0.6) c{M}. {F}inally, to establish bridges between the linkage groups of this and previous maps, 19 {SSR} markers were examined for polymorphism between the parents of this population. {W}e could only tentatively suggest a correspondence between four of our linkage groups and those of previous maps. {O}verall, {GBS} enabled us to quickly produce a genetic map with a density and uniformity of markers greater than previously published maps. {T}he availability of such a map will be useful for the identification of genomic regions associated with {S}triga resistance and other important agronomic traits.}, keywords = {{P}earl millet ; {G}enetic map ; {SNP} ; {G}enotyping-by-sequencing ; {AFRIQUE} {SUBSAHARIENNE} ; {INDE}}, booktitle = {}, journal = {{M}olecular {B}reeding}, volume = {35}, numero = {1}, pages = {art. 5 [10 p.]}, ISSN = {1380-3743}, year = {2015}, DOI = {10.1007/s11032-015-0212-x}, URL = {https://www.documentation.ird.fr/hor/fdi:010063913}, }
