@article{fdi:010063160, title = {{S}urvival of tissue balls from the coral {P}ocillopora damicornis {L}. exposed to cryoprotectant solutions}, author = {{F}euillassier, {L}ionel and {M}artinez, {L}. and {R}omans, {P}. and {S}ylvestre, {I}sabelle and {M}asanet, {P}. and {B}arth{\'e}l{\'e}my, {D}. and {E}ngelmann, {F}lorent}, editor = {}, language = {{ENG}}, abstract = {{I}n this study, the tolerance of tissue balls ({TB}s, 100-300 mu m in diameter) from the coral {P}ocillopora damicornis produced using mechanical excision to exposure to cryoprotectant ({CPA}) solutions was tested. {TB}s were treated for 20 min at room temperature with solutions of ethylene glycol ({EG}), methanol ({M}et), glycerol ({G}ly) or dimethyl sulfoxide ({M}e2{SO}) at concentrations between 1.0 and 4.5 {M}. {T}wo parameters were used to evaluate the survival of {TB}s following {CPA} treatment. {T}he {U}ndamaged {D}uration of {T}issue {B}alls (expressed in h) corresponded to the time period during which the membrane surface of {TB}s remained smooth and their motility was preserved. {T}issue {B}all {R}egression (expressed in mu m/h) corresponded to the size reduction of {TB}s over time. {TB}s tolerated exposure to all {CPA}s tested at the three lower concentrations employed (1.0 {M}, 1.5 {M} and 2.0 {M}). {N}o survival was achieved following exposure to a 4.5 {M} {CPA} solution. {A}t concentrations of 3.0 and 4.0 {M}, higher {U}ndamaged {D}uration of {T}issue {B}alls and lower {T}issue {B}all {R}egression were obtained following treatment with {EG} compared to the other three {CPA}s. {O}ur experiments show that {TB}s constitute a good experimental material to evaluate {CPA} toxicity on corals using large numbers of samples. {P}erforming preliminary experiments with {TB}s may allow reducing the number of tests carried out with less easily available coral forms such as planulae, thereby preserving larval stocks.}, keywords = {{T}issue ball ; {C}ryoprotectant ; {C}oral ; {C}ryopreservation ; {P}ocillopora damicornis}, booktitle = {}, journal = {{C}ryobiology}, volume = {69}, numero = {3}, pages = {376--385}, ISSN = {0011-2240}, year = {2014}, DOI = {10.1016/j.cryobiol.2014.08.009}, URL = {https://www.documentation.ird.fr/hor/fdi:010063160}, }
