@article{fdi:010063122, title = {{C}omparison of droplet-vitrification and {D}-cryoplate for cryopreservation of date palm ({P}hoenix dactylifera {L}.) polyembryonic masses}, author = {{S}alma, {M}. and {F}ki, {L}. and {S}ylvestre, {I}sabelle and {N}iino, {T}. and {E}ngelmann, {F}lorent}, editor = {}, language = {{ENG}}, abstract = {{I}n this work we tested the efficiency of two techniques, droplet-vitrification ({DV}) and dehydration ({D}) cryo-plate, for cryopreservation of polyembryonic masses ({PEM}s) of two date palm varieties, {S}okary and {S}ultany. {W}ith {DV}, recovery of non-precultured, cryopreserved {PEM}s was nil without treatment with the {PVS}2 vitrification solution, which contained 3.3 {M} glycerol + 2.4 {M} ethylene glycol + 0.4 {M} sucrose + 1.9 {M} dimethylsulfoxide. {F}ollowing {PVS}2 treatments between 15 and 120 min, it was comprised between 90.9-98.6% and 85.6-88.0% for varieties {S}okary and {S}ultany, respectively, {S}ucrose preculture (3 days, 0.5 {M}) led to 21.1% recovery of cryopreserved {PEM}s of variety {S}okary only without {PVS}2 treatment, and slightly reduced recovery in all other experimental conditions with both varieties, compared with noncryopreserved {PEM}s. {R}egrowth intensity of cryopreserved {PEM}s was generally lower with variety {S}ultany compared to variety {S}okary. {W}ith the {D} cryo-plate technique, no recovery of cryopreserved {PEM}s was achieved without sucrose preculture. {S}ucrose preculture (3 days, 0.5 {M}) had a positive effect on recovery of cryopreserved {PEM}s. {F}or variety {S}okary, the highest recovery (92.0-95.8%) was achieved for desiccation periods between 60 and 120 min. {R}ecovery was between 67.0 and 74.6% after desiccation periods of 90-120 min for variety {S}ultany. {W}ith the {D} cryo-plate technique, regrowth intensity of cryopreserved {PEM}s was higher with variety {S}okary compared to variety {S}ultany.}, keywords = {{D}ate palm ; {P}olyembryonic masses ; {C}ryopreservation ; {D}roplet-vitrification ; {D} cryo-plate}, booktitle = {}, journal = {{S}cientia {H}orticulturae}, volume = {179}, numero = {}, pages = {91--97}, ISSN = {0304-4238}, year = {2014}, DOI = {10.1016/j.scienta.2014.09.017}, URL = {https://www.documentation.ird.fr/hor/fdi:010063122}, }
