@article{fdi:010061991, title = {{S}ignificant impact of non-{B} {HIV}-1 variants genetic diversity in {G}abon on plasma {HIV}-1 {RNA} quantitation}, author = {{M}ouinga-{O}ndeme, {A}. and {M}abika-{M}abika, {A}. and {A}lalade, {P}. and {M}ongo, {A}. {D}. and {S}ica, {J}. and {L}i{\'e}geois, {F}lorian and {R}ouet, {F}.}, editor = {}, language = {{ENG}}, abstract = {{E}valuations of {HIV}-1 {RNA} viral load assays are lacking in {C}entral {A}frica. {T}he main objective of our study was to assess the reliability of {HIV}-1 {RNA} results obtained with three different assays for samples collected in {G}abon. {A} total of 137 plasma specimens were assessed for {HIV}-1 {RNA} using the {A}bbott {R}eal{T}ime {HIV}-1 ({R}) and {N}uclisens {HIV}-1 {E}asy{Q} ({R}) version 2.0 assays. {I}t included {HIV}-1 non-{B} samples (n = 113) representing six subtypes, 10 {CRF}s and 18 {URF}s from patients infected with {HIV}-1 and treated with antiretrovirals that were found {HIV}-1 {RNA} positive (>= 300 copies/ml) with the {G}eneric {HIV} viral load ({R}) assay; and samples (n = 24) from untreated individuals infected with {HIV}-1 but showing undetectable (<300 copies/ml) results with the {B}iocentric kit. {F}or samples found positive with the {G}eneric {HIV} viral load ({R}) test, correlation coefficients obtained between the three techniques were relatively low ({R} = 0.65 between {G}eneric {HIV} viral load ({R}) and {A}bbott {R}eal{T}ime {HIV}-1 ({R}), 0.50 between {G}eneric {HIV} viral load ({R}) and {N}uclisens {HIV}-1 {E}asy{Q} ({R}), and 0.66 between {A}bbott {R}eal{T}ime {HIV}-1 ({R}) and {N}uclisens {HIV}-1 {E}asy{Q} ({R})). {D}iscrepancies by at least one log(10) were obtained for 19.6%, 33.7%, and 20% of samples, respectively, irrespective of genotype. {M}ost of samples (22/24) from untreated study patients, found negative with the {B}iocentric kit, were also found negative with the two other techniques. {I}n {C}entral {A}frica, {HIV}-1 genetic diversity remains challenging for viral load testing. {F}urther studies are required in the same area to confirm the presence of {HIV}-1 strains that are not amplified with at least two different viral load assays. {J}. {M}ed. {V}irol. 86:52-57, 2014.}, keywords = {non-{B} subtypes/{CRF}s/{URF}s ; {A}frica ; {HIV}-1 {RNA} quantification ; {GABON}}, booktitle = {}, journal = {{J}ournal of {M}edical {V}irology}, volume = {86}, numero = {1}, pages = {52--57}, ISSN = {0146-6615}, year = {2014}, DOI = {10.1002/jmv.23770}, URL = {https://www.documentation.ird.fr/hor/fdi:010061991}, }
