@article{fdi:010061880, title = {{E}co-geographical diversity of cowpea bradyrhizobia in {S}enegal is marked by dominance of two genetic types}, author = {{W}ade, {T}. {K}. and {L}e {Q}u{\'e}r{\'e}, {A}ntoine and {L}aguerre, {G}. and {N}'{Z}oue, {A}. and {N}dione, {J}. {A}. and {D}orego, {F}. and {S}adio, {O}. and {N}doye, {I}. and {N}eyra, {M}arc}, editor = {}, language = {{ENG}}, abstract = {{T}he genetic diversity of native cowpea rhizobia originating from 60 sites across four eco-geographic zones in {S}enegal {W}as studied. {M}ore than 300 cowpea nodules were analyzed by {PCR}-{RFLP} of the 16{S}-23{S} r{DNA} {I}nter{G}enic {S}pacer region ({IGS}). {A}lignments of {IGS} sequences indicated that all genotypes were grouping within the {B}radyrhizobium genus. {T}he geographical distribution showed that apart from five {IGS} types, the others were specifically found in only one region. {T}he diversity was significantly higher in the {S}enegal {R}iver valley zone, which presents lower mean annual rainfalls and slightly alkaline soils. {I}nterestingly, two {IGS} types dominated the {S}enegalese rhizobial collection, one {IGS} type ({VI}) was found on more than half of the nodules collected in the northern {S}enegal {R}iver valley while another {IGS} type ({I}) was recovered from the great majority of nodules in the three other regions sampled. {T}wo representative strains from each of these two dominant types were isolated and further analyzed. {M}ulti {L}ocus {S}equence {A}nalyses using 6 housekeeping genes indicate that they belong to a new {B}radyrhizobium species closely related to {B}. yuanmingense. {P}hylogenetic analyses of 2 symbiotic genes nod{C} and nif{H} show that they are clustered with {B}. arachidis. {P}hysiological tests on these strains have shown that under laboratory conditions, the growth of the {IGS} type {VI} strains was slightly less affected by a higher osmotic strength in the medium and to alkaline p{H}, which corroborates the soil physico-chemical parameters.}, keywords = {{N}ative bradyrhizobia diversity ; {V}igna unguiculata ; 16{S}-23{S} r{DNA} {IGS} ; {H}ousekeeping genes ; nod{C} ; nif{H} ; {SENEGAL}}, booktitle = {}, journal = {{S}ystematic and {A}pplied {M}icrobiology}, volume = {37}, numero = {2}, pages = {129--139}, ISSN = {0723-2020}, year = {2014}, DOI = {10.1016/j.syapm.2013.10.002}, URL = {https://www.documentation.ird.fr/hor/fdi:010061880}, }
