@article{fdi:010061494, title = {{H}istocytological analysis of yam ({D}ioscorea alata) shoot tips cryopreserved by encapsulation-dehydration}, author = {{B}arraco, {G}. and {S}ylvestre, {I}sabelle and {C}ollin, {M}yriam and {E}scoute, {J}. and {L}artaud, {M}. and {V}erdeil, {J}. {L}. and {E}ngelmann, {F}lorent}, editor = {}, language = {{ENG}}, abstract = {{I}n this work, we performed qualitative and quantitative observations of the cytological changes occurring in cells of yam ({D}ioscorea alata) in vitro shoot tips cryopreserved using the encapsulation-dehydration ({E}-{D}) technique. {S}hoot tip osmoprotection for 24 h in 1.25 {M} sucrose medium induced drastic changes in cellular cytological features, including high plasmolysis in all three cellular areas studied, the external cell layer ({L}1), one to three ({L}1-3) and seven to nine ({L}7-9) cell layers from the surface of the meristematic dome, pyknotic nuclei in meristematic area cells and disappearance of nucleoli. {N}ucleus size decreased significantly in all cellular areas studied. {N}ucleocytoplasmic ratio decreased significantly in {L}1-3 and {L}7-9 cells. {N}uclear protein content increased, particularly in {L}1 and {L}1-3 cells. {A}fter physical dehydration, plasma membrane of numerous basal part cells was broken and intracellular soluble protein leakage was observed. {N}ucleus area and nucleocytoplasmic ratio decreased significantly in {L}7-9 cells. {O}ne week after cryopreservation, shoot tips showed regrowth and living cells had recovered their original morphology. {I}n all cellular areas studied, nuclei had retrieved their original staining and nucleoli were visible. {O}riginal nucleus area values were recovered in {L}1-3 and {L}1 cells. {T}he nucleocytoplasmic ratio retrieved its initial value in {L}1 cells but remained at levels observed after osmoprotection for {L}1-3 and {L}7-9 cells. {T}he nuclear protein content had retrieved its original level. {T}his investigation provided new insights in changes occurring in {D}. alata apices throughout an {E}-{D} protocol.}, keywords = {{D}ioscorea alata ; {S}hoot tip ; {C}ryopreservation ; {E}ncapsulation-dehydration ; {C}ytological features}, booktitle = {}, journal = {{P}rotoplasma}, volume = {251}, numero = {1}, pages = {177--189}, ISSN = {0033-183{X}}, year = {2014}, DOI = {10.1007/s00709-013-0536-5}, URL = {https://www.documentation.ird.fr/hor/fdi:010061494}, }
