@article{fdi:010061420, title = {{D}evelopment of real-time {RT}-{PCR} for the detection of low concentrations of {R}ift {V}alley fever virus}, author = {{M}aquart, {M}. and {T}emmam, {S}. and {H}eraud, {J}. {M}. and {L}eparc-{G}offart, {I}. and {C}etre-{S}ossah, {C}. and {D}ellagi, {K}oussay and {C}ardinale, {E}. and {P}ascalis, {H}erv{\'e}}, editor = {}, language = {{ENG}}, abstract = {{I}n recent years, {M}adagascar and the {C}omoros archipelago have been affected by epidemics of {R}ift {V}alley fever ({RVF}), however detection of {R}ift {V}alley fever virus ({RVFV}) in zebu, sheep and goats during the post epidemic periods was frequently unsuccessful. {T}hus, a highly sensitive real-time {RT}-{PCR} assay was developed for the detection of {RVFV} at low viral loads. {A} new {RVF} {SYBR} {G}reen {RT}-{PCR} targeting the {M} segment was tested on serum from different {RVF} seronegative ruminant species collected from {M}ay 2010 to {A}ugust 2011 in {M}adagascar and the {C}omoros archipelago and compared with a {RVF} specific quantitative real time {RT}-{PCR} technique, which is considered as the reference technique. {T}he specificity was tested on a wide range of arboviruses or other viruses giving {RVF} similar clinical signs. {A} total of 38 out of 2756 serum samples tested positive with the new {RT}-{PCR}, whereas the reference technique only detected 5 out of the 2756. {T}he described {RT}-{PCR} is an efficient diagnostic tool for the investigation of enzootic circulation of the {RVF} virus. {I}t allows the detection of low viral {RNA} loads adapted for the investigations of reservoirs or specific epidemiological situations such as inter-epizootic periods.}, keywords = {{R}ift {V}alley fever virus ; {D}iagnostic tool ; {S}emi-nested {RT}-{PCR} ; {I}nter-epidemic period ; {MAURITANIE} ; {MADAGASCAR} ; {COMORES} ; {MAYOTTE}}, booktitle = {}, journal = {{J}ournal of {V}irological {M}ethods}, volume = {195}, numero = {}, pages = {92--99}, ISSN = {0166-0934}, year = {2014}, DOI = {10.1016/j.jviromet.2013.10.001}, URL = {https://www.documentation.ird.fr/hor/fdi:010061420}, }
