@article{fdi:010060384, title = {{I}dentification of {G}lossina palpalis gambiensis specific salivary antigens : towards the development of a serologic biomarker of human exposure to tsetse flies in {W}est {A}frica}, author = {{D}ama, {E}. and {C}orn{\'e}lie, {S}ylvie and {S}omda, {M}. {B}. and {C}amara, {M}. and {K}ambire, {R}. and {C}ourtin, {F}abrice and {J}amonneau, {V}incent and {D}emettre, {E}. and {S}eveno, {M}. and {B}engaly, {Z}. and {S}olano, {P}hilippe and {P}oinsignon, {A}nne and {R}emou{\'e}, {F}ranck and {B}elem, {A}. {M}. {G}. and {B}ucheton, {B}runo}, editor = {}, language = {{ENG}}, abstract = {{T}he saliva of blood sucking arthropods contains a number of pharmacologically active compounds that induce an antibody response in exposed human individuals. {T}he objectives of the present study were (i) to assess the human {I}g{G} response directed against salivary antigens of {G}lossina palpalis gambiensis, the main vector of {T}rypanosoma brucei gambiense in {W}est {A}frica, as a biomarker of human tsetse contacts; and (ii) to identify specific salivary antigens. {I}mmune reactivity of human plasma collected within active human {A}frican trypanosomiasis ({HAT}) foci (coastal {G}uinea), historical foci where tsetse flies are still present ({S}outh-{W}est {B}urkina {F}aso) and a tsetse free area ({B}obo-{D}ioulasso, {B}urkina {F}aso), was measured by {ELISA} against whole saliva extracts. {I}n the active {HAT} foci and areas where tsetse flies were present in high densities, specific {I}g{G} responses were significantly higher (p < 0.0001) to those in {B}obo-{D}ioulasso or in {L}oropeni, where tsetse flies were either absent or only present at low densities. {F}urthermore, 2{D}-electrophoresis combined with mass spectrometry enabled to reveal that several antigens were specifically recognized by plasma from exposed individuals. {A}mong them, four salivary proteins were successfully identified ({A}da, 5'{N}uc, {A}g5 and {T}sgf1). {T}hese results represent a first attempt to identify {G}lossina salivary proteins or synthetic peptides to develop a standardized and specific biomarker of tsetse exposure in {W}est {A}frica.}, keywords = {{H}uman {A}frican trypanosomosis ; {G}lossina saliva ; {B}iomarker ; {E}xposure ; {AFRIQUE} {DE} {L}'{OUEST}}, booktitle = {}, journal = {{M}icrobes and {I}nfection}, volume = {15}, numero = {5}, pages = {416--427}, ISSN = {1286-4579}, year = {2013}, DOI = {10.1016/j.micinf.2013.03.001}, URL = {https://www.documentation.ird.fr/hor/fdi:010060384}, }
