@inproceedings{fdi:010007002, title = {{I}n vitro responses of senegalese adults {PBMC} to recombinant {MSP}1 19 antigens depends on expression systems and/or {P}lasmodium falciparum exposure}, author = {{G}arraud, {O}. and {D}iouf, {A}. and {L}ongacre, {S}. and {K}aslow, {D}.{C}. and {H}older, {A}.{A}. and {D}ieye, {A}. and {T}all, {A}. and {R}oussihon, {C}. and {T}rape, {J}ean-{F}ran{\c{c}}ois and {R}ogier, {C}. and {P}erraut, {R}. and {M}ercereau-{P}uijalon, {O}.}, editor = {}, language = {{ENG}}, abstract = {{T}o investigate the potential influence of antigen ({A}g) presentation on the immune response to {MSP}1 19, the 19 k{D}a {C}-terminal domain of the #{P}lasmodium falciparum$ {MSP}1 {A}g, 3 recombinant {MSP} 19 {A}gs expressed in baculovirus, #{E}. coli$ and #{S}accharomyces cerevisiae$ were used to stimulate in vitro peripheral blood mononuclear cells ({PBMCS}) from #{P}. falciparum$ immune adults with no recent history of clinical malaria. {D}onors were recruited in 2 senegalese villages ({D}ielmo and {N}diop), where malaria transmission is perennial and seasonal, respectively. {E}ach {PBMC} preparation was stimulated with a range of concentrations of the 3 recombinant {A}gs (cleaved from carrier molecules when appropriate). {M}ost subjects responded to at least 1 recombinant {A}g ({SI} > 3 in 8/10 at {D}ielmo and 14/15 at {N}diop). {T}he dose-dependent response to #{S}accharomyces cerevisiae$ {MSP}1 19 was similar in both villages. {I}n contrast, there was a 10-fold difference in the optimal concentration of the #{E}. coli$ product between the two villages. {B}oth the optimal dose and the concentration-dependence of the simulation by the baculovirus recombinant protein differed in each setting. {T}hese studies show that the 3 recombinant antigens differ in their capacity to stimulate lymphocyte proliferation, suggesting that protein structure (folding and/or glycosylation) affecting {A}g processing, is critical. {F}uthermore, these data suggest that the malaria transmission pattern influences the immune response to the various {MSP}1 19 recombinants.
