@article{PAR00014892, title = {{C}oxiella burnetii induces inflammatory interferon-like signature in plasmacytoid dendritic cells : a new feature of immune response in {Q} fever}, author = {{K}a, {M}. {B}. and {M}ezouar, {S}. and {B}en {A}mara, {A}. and {R}aoult, {D}idier and {G}higo, {E}. and {O}live, {D}. and {M}ege, {J}. {L}.}, editor = {}, language = {{ENG}}, abstract = {{P}lasmacytoid dendritic cells (p{DC}s) play a major role in antiviral immunity via the production of type {I} interferons ({IFN}s). {T}here is some evidence that p{DC}s interact with bacteria but it is not yet clear whether they are protective or contribute to bacterial pathogenicity. {W}e wished to investigate whether {C}oxiella burnetii, the agent of {Q} fever, interacts with p{DC}s. {T}he stimulation of p{DC}s with {C}. burnetii increased the expression of activation and migratory markers ({CD}86 and {CCR}7) as determined by flow cytometry and modulated gene expression program as revealed by a microarray approach. {I}ndeed, genes encoding for pro-inflammatory cytokines, chemokines, and type {I} {INF} were up-regulated. {T}he up-regulation of type {I} {IFN} was correlated with an increase in {IFN}-alpha release by {C}. burnetii-stimulated p{DC}s. {W}e also investigated p{DC}s in patients with {Q} fever endocarditis. {U}sing flow cytometry and a specific gating strategy, we found that the number of circulating p{DC}s was significantly lower in patients with {Q} fever endocarditis as compared to healthy donors. {I}n addition, the remaining circulating p{DC}s expressed activation and migratory markers. {A}s a whole, our study identified non-previously reported activation of p{DC}s by {C}. bumetil and their modulation during {Q} fever.}, keywords = {plasmacytoid dendritic cells ; {C}oxiella burnetii ; {Q} fever ; interferon ; infection}, booktitle = {}, journal = {{F}rontiers in {C}ellular and {I}nfection {M}icrobiology}, volume = {6}, numero = {}, pages = {70}, ISSN = {2235-2988}, year = {2016}, DOI = {10.3389/fcimb.2016.00070}, URL = {https://www.documentation.ird.fr/hor/{PAR}00014892}, }
