@article{PAR00014566, title = {{M}etagenomic analysis of antibiotic-induced changes in gut microbiota in a pregnant rat model}, author = {{K}han, {I}. and {A}zhar, {E}. {I}. and {A}bbas, {A}. {T}. and {K}umosani, {T}. and {B}arbour, {E}. {K}. and {R}aoult, {D}idier and {Y}asir, {M}.}, editor = {}, language = {{ENG}}, abstract = {{F}ood and {D}rug {A}dministration ({FDA}, {USA})-approved category {B} antibiotics are commonly prescribed to treat infections during pregnancy. {T}he aim of this study was to investigate antibiotic-induced changes in gut microbiota ({GM}) that occur during pregnancy. {T}he 16{S} r{RNA} amplicon deep-sequencing method was used to analyze the effect of category {B} antibiotics (azithromycin, amoxicillin and cefaclor) on {GM} during pregnancy using a rat model. {T}he {GM} composition was substantially modulated by pregnancy and antibiotics administration. {F}irmicutes, {B}acteroidetes, {P}roteobacteria,{C}hlamydiae, {A}ctinobacteria, and {C}yanobacteria were the dominant phyla. {A}ntibiotic treatment during pregnancy increased the relative abundance of {P}roteobacteria and reduced {F}irmicutes. {T}he genera {S}higella, {S}treptococcus, {C}andidatus {A}rthromitus, and {H}elicobacter were significantly (p < 0.05) more abundant during pregnancy. {A}ntibiotics significantly (p < 0.05) reduced the relative abundance of {L}actobacillus but increased that of {E}nterobacter. {T}here was a significant (p < 0.05) decrease in {L}actobacillussp., {L}actobacillus gallinarum and {L}actobacillus crispatus during pregnancy. {A}ntibiotictreatment reduced bacterial diversity; the lowest number of operational taxonomic units ({OTU}s) were detected in the cefaclor-treated groups. {A}ntibiotics significantly (p < 0.05) promoted weight gain during pregnancy, and increased relative abundance of {S}higella sonnei, {E}nterococcus hormaechei, and {A}cinetobacter sp. {GM} perturbations were accompanied by increases in {P}roteobacteria abundance and weight gain in pregnancy following antibiotic treatment.}, keywords = {gut microbiota ; azithromycin ; amoxicillin ; cefaclor ; pregnancy ; rats ; 16{S} r{RNA} gene}, booktitle = {}, journal = {{F}rontiers in {P}harmacology}, volume = {7}, numero = {}, pages = {art. 104}, ISSN = {1663-9812}, year = {2016}, URL = {https://www.documentation.ird.fr/hor/{PAR}00014566}, }
