@article{PAR00014305, title = {{A} chlorhexidine-agar plate culture medium protocol to complement standard broth culture of {M}ycobacterium tuberculosis}, author = {{A}smar, {S}. and {C}hatellier, {S}. and {M}irande, {C}. and van {B}elkum, {A}. and {C}anard, {I}. and {R}aoult, {D}idier and {D}rancourt, {M}.}, editor = {}, language = {{ENG}}, abstract = {{T}he culture of {M}ycobacterium tuberculosis using parallel inoculation of a solid culture medium and a liquid broth provides the gold standard for the diagnosis of tuberculosis. {H}ere, we evaluated a chlorhexidine decontamination-{MOD}9 solid medium protocol versus the standard {NALC}-{N}a{OH}-{B}actec 960 {MGIT} protocol for the diagnosis of pulmonary tuberculosis by culture. {T}hree-hundred clinical specimens comprising 193 sputa, 30 bronchial aspirates, 10 broncho-alveolar lavages, 47 stools, and 20 urines were prospectively submitted for the routine diagnosis of tuberculosis. {T}he contamination rates were 5/300 (1.7%) using the {MOD}9 protocol and 17/300 (5.7%) with the {B}actec protocol, respectively ({P} < 0.05, {F}isher exact test). {O}f a total of 50 {M}ycobacterium isolates (48 {M}. tuberculosis and two {M}ycobacterium abscessus) were cultured. {O}ut of these 50, 48 (96%) isolates were found using the {MOD}9 protocol versus 35 (70%) when using the {B}actec protocol ({P} < 05, {F}isher exact test). {T}he time to positivity was 10.1 +/- 3.9 days versus 14.7 +/- 7.3 days, respectively, ({P} < 0.05, {S}tudent's t-test). {T}hese data confirmed the usefulness of parallel inoculation of a solid culture medium with broth for the recovery of {M}. tuberculosis in agreement with current recommendations. {M}ore specifically, chlorhexidine decontamination and inoculation of the {MOD}9 solid medium could be proposed to complement the standard {B}actec 960 {MGIT} broth protocol.}, keywords = {{M}ycobacterium tuberculosis ; diagnosis ; decontamination ; chlorhexidine ; {MOD}9 ; {B}actec 960 {MGIT} ; {N}-acetyl-cysteine-sodium chloride}, booktitle = {}, journal = {{F}rontiers in {M}icrobiology}, volume = {7}, numero = {}, pages = {art. 30}, ISSN = {1664-302{X}}, year = {2016}, DOI = {10.3389/fmicb.2016.00030}, URL = {https://www.documentation.ird.fr/hor/{PAR}00014305}, }
