@article{PAR00013855, title = {{I}solation of new {B}razilian giant viruses from environmental samples using a panel of protozoa}, author = {{D}ornas, {F}. {P}. and {K}halil, {J}. {Y}. {B}. and {P}agnier, {I}. and {R}aoult, {D}idier and {A}brahao, {J}. and {L}a {S}cola, {B}.}, editor = {}, language = {{ENG}}, abstract = {{T}he {M}egavirales are a newly described order capable of infecting different types of eukaryotic hosts. {F}or the most part, the natural host is unknown. {S}everal methods have been used to detect these viruses, with large discrepancies between molecular methods and co-cultures. {T}o isolate giant viruses, we propose the use of different species of amoeba as a cellular support. {T}he aim of this work was to isolate new {B}razilian giant viruses by comparing the protozoa {A}canthamoeba castellani {I}, {A}. polyphaga, {A}. griffini, and {V}ermamoeba vermiformis ({VV}) as a platform for cellular isolation using environmental samples. {O}ne hundred samples were collected from 3 different areas in {S}eptember 2014 in the {P}ampulha lagoon of {B}elo {H}orizonte city, {M}inas {G}erais, {B}razil. {PCR} was used to identify the isolated viruses, along with hemacolor staining, labelling fluorescence and electron microscopy. {A} total of 69 viruses were isolated. {T}he highest ratio of isolation was found in {A}. polyphaga (46.38%) and the lowest in {W} (0%). {M}imiviruses were the most frequently isolated. {O}ne {M}arseillevirus and one {P}andoravirus were also isolated. {W}ith {B}razilian environmental samples, we demonstrated the high rate of lineage {A} mimiviruses. {T}his work demonstrates how these viruses survive and circulate in nature as well the differences between protozoa as a platform for cellular isolation.}, keywords = {{M}imivirus ; {M}arseillevirus ; {P}andoravirus ; {M}egavirales ; giant virus ; isolation ; acanthamoeba ; {BRESIL}}, booktitle = {}, journal = {{F}rontiers in {M}icrobiology}, volume = {6}, numero = {}, pages = {art. 1086 [9 p.]}, ISSN = {1664-302{X}}, year = {2015}, DOI = {10.3389/fmicb.2015.01086}, URL = {https://www.documentation.ird.fr/hor/{PAR}00013855}, }
