@article{PAR00013603, title = {{P}ithovirus sibericum, a new bona fide member of the "{F}ourth {TRUC}" club}, author = {{S}harma, {V}. and {C}olson, {P}. and {C}habrol, {O}. and {P}ontarotti, {P}. and {R}aoult, {D}idier}, editor = {}, language = {{ENG}}, abstract = {{N}ucleocytoplasmic large {DNA} viruses, or representatives of the proposed order {M}egavirales, include giant viruses of {A}canthamoeba that were discovered over the last 12 years and are bona fide microbes. {P}hylogenies based on a few genes conserved amongst these megaviruses and shared by microbes classified as {E}ukarya, {B}acteria, and {A}rchaea, allowed for delineation of a fourth monophylogenetic group or "{TRUC}" ({T}hings {R}esisting {U}ncompleted {C}lassification) composed of the {M}egavirales representatives. {A} new {M}egavirales member named {P}ithovirus sibericum was isolated from a >30,000-year-old dated {S}iberian permafrost sample. {T}his virion is as large as recently described pandoraviruses but has a genome that is approximately three to four times shorter. {O}ur objective was to update the classification of {P}. sibericum as a new member of the "{F}ourth {TRUC}" club. {P}hylogenetic trees were constructed based on four conserved ancient genes and a phyletic analysis was concurrently conducted based on the presence/absence patterns of a set of informational genes from members of {M}egavirales, {B}acteria, {A}rchaea, and {E}ukarya. {P}hylogenetic analyses based on the four conserved genes revealed that {P}. sibericum is part of the fourth {TRUC} composed of {M}egavirales members, and is closely related to the families {M}arseilleviridae and {A}scoviridae/{I}ridoviridae. {A}dditionally, hierarchical clustering delineated four branches, and showed that {P}. sibericum is part of this fourth {TRUC}. {O}verall, phylogenetic and phyletic analyses using informational genes clearly indicate that {P}. sibericum is a new bona fide member of the "{F}ourth {TRUC}" club composed of representatives of {M}egavirales, alongside {B}acteria, {A}rchaea, and {E}ukarya.}, keywords = {giant virus ; {P}ithovirus sibericum ; {M}egavirales ; nucleocytoplasmic large ; {DNA} viruses ; informational genes ; {TRUC} ; domains of life ; phylogeny}, booktitle = {}, journal = {{F}rontiers in {M}icrobiology}, volume = {6}, numero = {}, pages = {art. 722}, ISSN = {1664-302{X}}, year = {2015}, DOI = {10.3389/fmicb.2015.00722}, URL = {https://www.documentation.ird.fr/hor/{PAR}00013603}, }
