@article{PAR00012921, title = {{D}etection of {R}ickettsia spp in ticks by {MALDI}-{TOF} {MS}}, author = {{Y}ssouf, {A}. and {A}lmeras, {L}. and {T}erras, {J}. and {S}ocolovschi, {C}. and {R}aoult, {D}idier and {P}arola, {P}.}, editor = {}, language = {{ENG}}, abstract = {{B}ackground {M}atrix {A}ssisted {L}aser {D}esorption/{I}onization {T}ime-of-{F}light {M}ass {S}pectrometry ({MALDI}-{TOF} {MS}) has been shown to be an effective tool for the rapid identification of arthropods, including tick vectors of human diseases. {M}ethodology/{P}rincipal {F}indings {T}he objective of the present study was to evaluate the use of {MALDI}-{TOF} {MS} to identify tick species, and to determine the presence of rickettsia pathogens in the infected {T}icks. {R}hipicephalus sanguineus and {D}ermacentor marginatus {T}icks infected or not by {R}. conorii conorii or {R}. slovaca, respectively, were used as experimental models. {T}he {MS} profiles generated from protein extracts prepared from tick legs exhibited mass peaks that distinguished the infected and uninfected {T}icks, and successfully discriminated the {R}ickettsia spp. {A} blind test was performed using {T}icks that were laboratory-reared, collected in the field or removed from patients and infected or not by {R}ickettsia spp. {A} query against our in-lab arthropod {MS} reference database revealed that the species and infection status of all {T}icks were correctly identified at the species and infection status levels. {C}onclusions/{S}ignificance {T}aken together, the present work demonstrates the utility of {MALDI}-{TOF} {MS} for a dual identification of tick species and intracellular bacteria. {T}herefore, {MALDI}-{TOF} {MS} is a relevant tool for the accurate detection of {R}ickettsia spp in {T}icks for both field monitoring and entomological diagnosis. {T}he present work offers new perspectives for the monitoring of other vector borne diseases that present public health concerns.}, keywords = {}, booktitle = {}, journal = {{P}los {N}eglected {T}ropical {D}iseases}, volume = {9}, numero = {2}, pages = {e0003473 [16 p.]}, ISSN = {1935-2735}, year = {2015}, DOI = {10.1371/journal.pntd.0003473}, URL = {https://www.documentation.ird.fr/hor/{PAR}00012921}, }
