@article{PAR00011193, title = {{F}irst isolation of a {M}arseillevirus in the diptera {S}yrphidae {E}ristalis tenax}, author = {{B}oughalmi, {M}. and {P}agnier, {I}. and {A}herfi, {S}. and {C}olson, {P}. and {R}aoult, {D}idier and {L}a {S}cola, {B}.}, editor = {}, language = {{ENG}}, abstract = {{O}bjective: {G}iant viruses and amoebae are common in freshwater, where they can coexist with various insects. {W}e screened insect larvae to detect giant viruses using a high-throughput method. {M}ethods: {W}e analyzed 86 {E}ristalis tenax larvae obtained from stagnant water reservoirs in {T}unisia. {T}he larvae were decontaminated and then dissected to remove internal parts for coculture with {A}canthamoeba polyphaga. {G}enome sequencing of isolated viruses was performed on a 454 {R}oche instrument, and comparative genomics were performed. {R}esults: {O}ne {M}arseillevirus, named {I}nsectomime virus, was isolated. {T}he genome assembly generated two scaffolds, which were 382,776 and 3,855 bp in length. {A}mong the 477 identified predicted proteins, the best hit for 435 of the identified proteins was a {M}arseillevirus or {L}ausannevirus protein. {T}unisvirus was the most closely related to {I}nsectomime, with 446 orthologs. {O}ne {I}nsectomime protein shared with {L}ausannevirus and {T}unisvirus showed the highest similarity with a protein from an aphid. {C}onclusion: {T}he isolation of a {M}arseillevirus from an insect expands the diversity of environments in which giant viruses have been isolated. {T}he coexistence of larvae and giant viruses in stagnant water may explain the presence of the giant virus in the larva internal structures. {T}his study illustrates the putative role of amoeba in lateral gene transfer not only between the organisms it phagocytoses, but also between organisms living in the same environment.}, keywords = {{G}iant virus ; {M}arseillevirus ; {M}arseilleviridae ; {E}ristalis tenax}, booktitle = {}, journal = {{I}ntervirology}, volume = {56}, numero = {6}, pages = {386--394}, ISSN = {0300-5526}, year = {2013}, DOI = {10.1159/000354560}, URL = {https://www.documentation.ird.fr/hor/{PAR}00011193}, }
